Sometime after Simba becomes King of the Pride Lands, he and Nala have a daughter named Kiara and are expecting another child. They set off to an oasis where Nala can give birth, while Simba asks Timon and Pumbaa to keep an eye on Kiara. Rafiki soon visits them and decides to share the story of his grandfather Mufasa and his treacherous younger brother Scar, with Timon and Pumbaa making colorful comments to each other.

A long time ago, Mufasa was born into a small family consisting of himself and his parents, Masego and Avia, who believed in a mythical land called Melele. A flood hits and Mufasa's fear prevents him from reaching Avia; he was swept into the waters where he met a royal cub named Taka, who saved him from crocodiles before Taka's mother, Queen Ishii, intervened. Ishii reluctantly accepts Mufasa as his son, but Taka's father, King Obasi, criticizes Taka for taking in an outsider. Mufasa claims he can serve the pride and races with Taka to prove his speed. Mufasa falls behind due to exhaustion, but Taka admits defeat so Mufasa can rejoin the pride.

As Mufasa and Taka grow, they form a strong brotherly bond. While Ishii teaches Mufasa how to hunt, the land is attacked by two white lions that attack Mufasa and Ishii, but Mufasa kills one of them while the other flees. Taka sees the attack but backs away in fear. The survivor is returned to Kyros, leader of a pride of white lions called "The Outsiders", composed of lions exiled for their white fur, and father of the lion killed by Mufasa; Kyros and his sisters Aqua and Amara swear revenge. The strangers kill Obasi, Ishi, and their pride before pursuing Mufasa and Taka, who escape into the river.

When Mufasa and Taka arrive on Earth, they encounter a wandering lioness named Sarabi, her friend Zazu, and a young Rafiki. Rafiki tells them that he is going to Melili to find a new home, and the group follows him there. Romantic feelings develop between Taka and Sarabi as strangers continue to pursue them. Sarabi uses bees to cause a stampede of elephants to distract the strangers, allowing the group to escape. She falls and is injured, forcing Mufasa to save her, but he lies and tells a recovering Sarabi that Taka saved her.

The group hides in a snowy mountain range to recover. Sarabi tells Mufasa that she knows he saved her, and they soon fall in love. A jealous Taka watches this in secret, meets with Kyros and his pride, and offers Kyros revenge on Mufasa for his son's death. The next day, the group arrives at Melili, a fertile oasis, and Taka secretly leaves signs for the outsiders to follow.

Rafiki claims Mufasa is his brother before the outsiders come to attack him. Mufasa learns of Taka's betrayal while fighting Kiros. Despite this, Mufasa rallies the animals of Melili to fight the outsiders. Kiros forces Mufasa into a cave. Taka sees his exhausted brother and realizes what he has done, intervenes with remorse, and during the struggle, Kyros hits Taka in the eye with his paw, leaving a scar. Rafiki causes the cave to collapse, causing it to flood. Aqua and Amara are killed in the collapse while attempting to kill Sarabi. Kyros attempts to drown Mufasa, but a falling rock hits him and kills Kyros as Mufasa swims to safety. Taka is tempted to drown his brother but helps him out of the water.

The flood recedes and Mufasa and Sarabi exit the cave together as the animals celebrate their victory. Rafiki calls the couple the king and queen. Mufasa soon meets Afia, who tells him that Masego died in the previous flood. Mufasa confronts Taka about his betrayal and allows him to stay, but as he refuses to refer to Taka by his name, Taka renames himself "Scar". Mufasa then climbs the newly formed Pride Rock and cheers in triumph.

In the present, Kiara roars atop Pride Rock as her grandfather's spirit towers over her. She and her friends soon meet Simba to meet his newborn brother.
